Western Blot: Tubulin Antibody (YL1/2) [NB600-506] - Total protein from human HeLa, mouse 3T3, rat PC12 and African green monkey Cos7 cell lines was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with a 1:6000 dilution of anti-Tubulin (YL 1/2) in 1% non-fat milk in TBST and detected with an anti-rat HRP secondary antibody using chemiluminescence.
Immunocytochemistry/Immunofluorescence: Tubulin Antibody (YL1/2) [NB600-506] - HeLa cells were fixed and permeabilized for 10 minutes using -20C MeOH. The cells were incubated with anti-Tubulin (YL1/2) at a 1:200 dilution overnight at 4C and detected with an anti-rat Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Tubulin alpha-1A chain (TUBA1A), also known as alpha-tubulin 3, tubulin B-alpha-1, B-ALPHA-1, tubulin alpha-3 chain, TUBA3, and LIS3, is a member of the tubulin family. Tubulin is the primary constituent of microtubules. Tubulin binds two moles of GTP, one at an exchangeable site on the beta chain and one at a non-exchangeable site on the alpha-chain. The TUBA1A gene is one of three alpha tubulin genes located on chromosome 12q. TUBA1A is found on the cytoskeleton and is expressed predominantly in morphologically differentiated neurologic cells. Mutations to the TUBA1A gene cause a neurological condition called lissencephaly type 3 (LIS3). As a result of defective neuronal migration, LIS3 is characterized by microcephaly, mental retardation, and early-onset epilepsy (PMID: 22264709). The tubulin clone (YL1/2) specifically binds tyrosylated Tubulin (Tyr-Tubulin) (PMID: 6415068).
