The pro-alpha 1 subunit of type 1 collagen is encoded by the COL1A1 gene. The collagen type 1 protein is a triple helix composed by one alpha 2 chain and two alpha 1 chains. Type 1 collagen is abundant in bone, dermis, tendon and cornea tissues and is the fibril forming collagen found in the majority of connective tissues. A type of skin tumor called dermatofibrosarcoma protuberans has been linked to reciprocal translocations between the 17th and 22nd chromosomes where the COL1A1 gene is located. Alternate polyadenylation signals result in two transcripts for the collagen type 1 gene. COL3A1 is an important paralog of this gene. Studies using the collagen I alpha 1 antibody show that COL-I degradation is required for tumor growth in certain cancer types where a MT1-MMP complex acts to destroy collagen(1). In a study conducted at the University of Minnesota the collagen I alpha 1 antibody was used to look at the efficacy of using HBOECs (human blood outgrowth endothelial cells) to form tissue-engineered vascular grafts by examining collagen deposition on bioartificial tissue that was either seeded with HBOECs or unseeded(2). Analysis of collagen structure and deposition in tissues using the collagen I alpha 1 antibody has shown that this collagen I alpha 1 antibody is a useful tool in both cancer and wound healing research areas.
