	Western Blot: FoxP3 Antibody [NB600-242] - (1ug/ml) staining of Human Muscle (A), (2ug/ml) MOLT4 (B) and (1ug/ml) negative control Pancreas (C) lysate (35ug protein in RIPA buffer). Detected by chemiluminescence.
	Immunocytochemistry/Immunofluorescence: FoxP3 Antibody [NB600-242] - Analysis of paraformaldehyde fixed U2OS cells, permeabilized with 0.15% Triton. Primary incubation 1hr (10ug/ml) followed by Alexa Fluor 488 secondary antibody (4ug/ml), showing strong localization to nucleoplasm. The nuclear stain is DAPI (blue). Negative control: Unimmunized goat IgG (10ug/ml) followed by Alexa Fluor 488 secondary antibody (4ug/ml).
	Flow Cytometry: FoxP3 Antibody [NB600-242] - Analysis of paraformaldehyde fixed Jurkat cells (blue line), permeabilized with 0.5% Triton. Primary incubation 1hr (10ug/ml) followed by Alexa Fluor 488 secondary antibody (4ug/ml). IgG control: Unimmunized goat IgG (black line) followed by Alexa Fluor 488 secondary antibody.
	Immunocytochemistry/Immunofluorescence: FoxP3 Antibody [NB600-242] - (Treg) human blood cells gathered by cytospin and detected by FITC (A) and in phase contrast (B).

Background

FOXP3 (a 381 amino acid protein) is a member of the forkhead/winged-helix family of transcriptional regulators and is highly conserved across mammals. FOXP3 is essential for normal immune homeostasis. FOXP3 is stably and constitutively expressed at a high level in CD25 + CD4 positive regulatory T cells, a low level in CD4-positive/CD25-negative cells, and is absent in CD4-negative/CD8-positive T cells. FOXP3 may be a master regulatory gene and a more specific marker of regulatory T cells than other T cells.
