	Immunohistochemistry-Paraffin: TLR7 Antibody [NB100-56588] - IHC analysis of formalin fixed paraffin-embedded (FFPE) human brain using TLR7 antibody at 1:20 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Neuronal staining of TLR7 was observed. Staining was performed by Histowiz.

Background

The Toll-like receptor (TLR) family in mammal comprises a family of transmembrane proteins characterized by multiple copies of leucine rich repeats in the extracellular domain and IL-1 receptor motif in the cytoplasmic domain. Like its counterparts in Drosophila, TLRs signal through adaptor molecules and could constitute an important and unrecognized component of innate immunity in humans. The TLR family is a phylogenetically conserved mediator of innate immunity that is essential for microbial recognition. TLRs characterized so far activate the MyD88/interleukin-1 receptor-associated kinase (IRAK) signaling pathway. Ten human homologs of TLRs (TLR1-10) have been described. Stimulation of the NFkB signaling pathway by TLR7 suggests that it plays a role in immune response.
