 | Detection of Human and Mouse S100A6 by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line, RAW 264.7 mouse monocyte/macrophage cell line, and NIH‑3T3 mouse embryonic fibroblast cell line. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human S100A6 Monoclonal Antibody (Catalog # MAB7467) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for S100A6 at approximately 10 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1. |

 | S100A6 in Human Kidney. S100A6 was detected in immersion fixed paraffin-embedded sections of human kidney using Mouse Anti-Human S100A6 Monoclonal Antibody (Catalog # MAB7467) at 15 µg/mL overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counterstained with hematoxylin (blue). | background_content | Background: S100A6 S100A6, also called PRA (prolactin receptor-associated protein) and calcyclin, is a 10 kDa member of the S100 family of calcium-binding proteins. Human S100A6 is 90 amino acids (aa) in length and contains two calcium-binding EF-hand domains (aa 12 - 47 and 48 - 83). Intracellularly, S100A6 will both noncovalently homodimerize and heterodimerize with S100B plus SGT1. Extracellularly, it is secreted via a noncanonical pathway and binds to RAGE, inducing apoptosis. It is expressed by neurons, endothelium, fibroblasts and glandular epithelia. Human S100A6 shares 95% aa sequence identity with mouse and rat S100A6. This antibody detects mouse S100A6 in Western Blots. Mouse cross-reactivity has not been tested in Immunohistochemistry |
