APPLICATION

Application Note

ELISA: Use at a concentration of 0.1 - 1.0 μg/ml. IHC: Use at a dilution of 1/100 - 1/400. WB: Use at a concentration of 0.5 - 2.0 μg/ml. Predicted molecular weight: 42 kDa. The formalin-fixed tissues were boiled in 10 mM citrate buffer, pH 6.0 for 10 min, followed by cooling at RT for 20 min. Optimal dilutions/concentrations should be determined by the end user.

Specificity/Sensitivity	This antibody stains smooth muscle cells in vessel walls, gut wall, and myometrium. Myoepithelial cells in breast and salivary gland are also stained. It reacts with tumors arising from smooth muscles and myoepithelial cells. The other actins, such as beta- and gamma-cytoplasmic, striated muscle, and myocardium are not stained by this antibody.

Background	The protein encoded by this gene belongs to the actin family of proteins, which are highly conserved proteins that play a role in cell motility, structure and integrity. Alpha, beta and gamma actin isoforms have been identified, with alpha actins being a major constituent of the contractile apparatus, while beta and gamma actins are involved in the regulation of cell motility. This actin is an alpha actin that is found in skeletal muscle. Defects in this gene cause aortic aneurysm familial thoracic type 6. Multiple alternatively spliced variants, encoding the same protein, have been identified. [provided by RefSeq, Nov 2008]
