 | Detection of Human, Mouse, and Rat GRK7 by Western Blot. Western blot shows lysates of A431 human epithelial carcinoma cell line, BJAB human Burkitt's lymphoma cell line, BaF3 mouse pro-B cell line, and Rat‑2 rat embryonic fibroblast cell line. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human/Mouse/Rat GRK7 Monoclonal Antibody (Catalog # MAB4700) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for GRK7 at approximately 62 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1. |

| background_content | Background: GRK7 G protein-coupled receptor kinases (GRKs) are important modulators of G protein-coupled receptor (GPCR) signaling. Receptor phosphorylation by specific GRKs plays a key role in triggering rapid desensitization. The GRK family consists of 7 isoforms that share a central catalytic domain with homology to other serine/threonine kinases. The catalytic domain is flanked by an amino-terminal RGS domain of 183-188 amino acids and a carboxyl-terminus of variable length. GRK1 and GRK7 are members of the rhodopsin kinase or visual GRK subfamily. GRK7 is expressed in the cone outer segments of the retina and is able to phosphorylate rhodopsin in a light-dependent manner. In addition to its retina expression, GRK7 has been detected in other tissues, including brain, olfactory bulb, liver, lung and pancreas. |
