細胞表面レセプター　in situ プロファイリング用キット MiSEP Receptor Profiling Kit

Luminex^(R) xMAP^(R) システムを用いて，ヒト細胞の表面で発現しているレセプターを in situ でプロファイリングできるキットです。細胞表面に発現している複数のエピトープを認識する抗体カクテルを用いることにより，1 回のアッセイで多種類のレセプターを同時に検出することができます。細胞の剥離が不要なため，細胞表面レセプターの発現に影響を与えず，信頼性の高い結果が得られます。

接着細胞は剥離が不要なため，剥離による人工的な発現変化が引き起こされません。
接着細胞および浮遊細胞のいずれにも使用できます。
抗体は，標的レセプターの細胞外ドメイン（ECD）をエピトープとするものを使用しています。
フローサイトメトリーよりも高感度かつ高い S/N 比で検出できます。
キットには，内部標準としてトランスフェリンレセプター（TfR）が含まれており，多検体間での補正が容易に行えます。
1 回のアッセイで複数のレセプターを同時に解析できます。
使用する細胞数は接着細胞で 2.5 × 10⁵ 個／ウェル，浮遊細胞で≧ 1 × 10⁶ 個／ウェルのため，貴重な試料を無駄にしません。
複合体を形成するタンパク質の検出も可能です。

操作法概略

抗体カクテルと試料をインキュベートし，細胞表面のエピトープと抗体を結合させる。
洗浄し，未結合の抗体を取り除く。
エピトープに結合した抗体を細胞から溶出する。
溶出した抗体を回収し，Bead set およびLuminex^(R) xMAP^(R) システムを使用して定量する。
