R&D Systems now offers Recombinant Human IL-32 alpha (Catalog # 3040‑IL). IL-32 is a secreted glycoprotein that is up-regulated by inflammatory stimulation of monocytes, natural killer cells, epithelial cells, and pancreatic myofibroblasts. It promotes the expression of pro-inflammatory molecules, such as TNF-alpha, IL-1, IL-6, and CXCL8/IL-8. The alpha isoform of IL-32 is one of six potential splice variants. It lacks a portion of the putative signal peptide and 57 amino acids from the C-terminal region of the longest IL-32 isoform. Cleavage of IL-32 alpha by neutrophil-derived Proteinase 3 releases an N-terminal fragment with increased potency for stimulating CXCL2/MIP-2 and CXCL8/IL-8 expression. IL-32 is highly expressed in a number of pathologicalconditions, including inflammatory bowel disease, rheumatoid arthritis, chronic pancreatitis, and pancreatic cancer. | | IL-32 alpha Induces TNF-alpha Secretion in RAW264.7 Mouse Macrophage Cells. RAW264.7 mouse macrophage cells were treated with increasing concentrations of Recombinant Human IL-32 alpha (Catalog # 3040-IL) in the presence of 2 µg/mL muramyl dipeptide (MDP). The concentration of TNF-alpha was measured 24 hours later using the Mouse TNF-alpha Quantikine® ELISA Kit (Catalog # MTA00). |
